Once formed, 1a, 25 OH ; 2D3 regulates serum concentrations of Ca2 and phosphorus by increasing the efficiency of Ca2 and phosphorus absorption from the intestine and by mobilizing Ca2 stores from bone. 1a, 25 OH ; 2D3 also downregulates the expression of PTH and 1a-OHase in a feedback mechanism that regulates the synthesis of 1a, 25 OH ; 2D3. Ultimately, vitamin D maintains Ca2 and phosphorus levels within the normal serum range, to sustain a variety of metabolic functions, physiologic functions and bone health. Many effective therapeutic agents exhibit effects that are different from their intended primary mode of action. Antibiotics such as doxycycline and erythromycin A are no exception. They also display anti-inflammatory activity. Using LA4 murine lung alveolar epithelial cells, effects of doxycycline and erythromycin A on inducible NO synthase iNOS ; NO production as well as iNOS protein and mRNA production were investigated. Induction of iNOS was accomplished by treatment with cytomix TNF- , IL-1 , and IFN- each at 5 ng Production of NO or iNOS was not detected in controls with or without erythromycin A. In the presence of cytomix, erythromycin A did not decrease NO, nitrite, iNOS protein, or mRNA production. In contrast, doxycycline caused a dose-dependent decrease in NO, nitrite, iNOS protein, and mRNA production in cytomix-treated cells. Doxycycline at 30 g produced a 90% decrease in nitrite and NO production and a 52% decrease in iNOS mRNA transcription compared with cytomix treatment alone. Actinomycin D treatment suggests that doxycycline decreases stability of iNOS mRNA in cytomix-treated cells. To determine a mechanism for the decrease in iNOS expression, NF- B and AP-1 transcription regulatory systems and p38 MAPK were examined. Doxycycline treatment gave no statistically significant change in NF- B activation but did decrease p38 MAPK protein in cytomix-treated cells by 50%, suggesting that p38 MAPK may be responsible for stabilization of iNOS mRNA. These results demonstrate that doxycycline decreases NO production from iNOS by destabilization of iNOS mRNA via decreased expression of p38 MAPK. The Journal of Immunology, 2006, 176: 567572. he common obstructive airway disorders such as asthma, chronic bronchitis, diffuse panbronchiolitis, and bronchiectasis are all invariably associated with chronic airway inflammation 13 ; . Despite treatment with anti-inflammatory therapies, these disorders are often progressive, suggesting that the current anti-inflammatory therapies are inadequate. Macrolides and tetracyclines are bacterial and fungal secondary metabolites that possess antibacterial action but also exhibit distinct and separate anti-inflammatory actions. These antibacterial compounds have been known to reduce inflammation associated with a variety of inflammatory diseases since the 1970s 4, 5 ; . Among these compounds are erythromycin A and many of its derivatives such as azithromycin, clarithromycin, and roxithromycin, as well as tetracycline and several related compounds such as doxycycline. These compounds have exhibited anti-inflammatory activity against a variety of inflammatory airway disorders 5 ; . However, concern has been raised about development of microbial antibiotic resistance limiting macrolide usefulness and suggesting a need for alternative anti-inflammatory therapies. One marker of inflammation has been measurement of NO, a gas produced by lung epithelial cells, which has been shown to be elevated in several inflammatory airway conditions 6 ; . We hypothesized that doxycycline possesses potent anti-inflammatory properties in murine lung epithelial cells, leading to a reduction of and tegaserod.

Background: Several cell types in the blood produce microvesicles MV under activating conditions. Due to an interaction of Pplatelets and Monocytes under such conditions we were interested in the interactions of their vesicles PMV and MMV ; , the stability of their conjugates and the formation kinetics of MV and conjugates. Methods: A native plasma sample was obtained by centrifugation of citrated whole blood WB ; for 5 min at 5, 000 x g. MV PMV, MMV ; and their conjugates were generated by incubating samples of this WB with collagen 5 g ml ; LPS 1g ml ; or both or without activator for 4 hours at 37 under gentle agitation. At several time points 1ml WB was separated. After preparing PPP, the samples were divided and one part was incubated with EDTA. MV were stained with fluorescent antiCD42a and anti-CD14 and analyzed by two color flow cytometry. MV and conjugate numbers were determined by counting them at a constant acquisition time of 20s and subsequently analyzed using logical gates. Results: The number of detectable MV and conjugates is increasing from 50000 to ca. 100000 after 4h incubation in all samples. The presence of an activator was visible only in the first 2h incubation, especially in the presence of collagen the numbers increased in this time almost to the end level. A difference between samples with, for example, roxithromucin 300 mg.

Tesunate, and thus, increase its cerebral concentration. SwissWebster female mice infected with the RH strain of T. gondii and treated subcutaneously with different daily doses of arteether showed more protection compared with mice treated orally with roxithromycin.9 These data were later confirmed by Brun-Pascaud and others, 10 who showed that after 5 weeks of immunosuppression, rats inoculated with the RH strain of T. gondii and treated with artemether at doses of 18 and 100 mg kg died within 8 days. These mice had been treated with a total dose of 500 mg over a 5-day period and treatment was well tolerated. Cooke and others16 showed that 1 hr after a 900 mg kg oral dose was given, a concentration of 6 g was observed in rat brain tissue. The total dose in our study was less than this value, but the number of cerebral cysts decreased by 59% in treated animals compared with the controls. Thus, this low cerebral dose displayed a certain efficacy. Our microscopic study showed that cysts treated with the drugs generally appeared unchanged, but an alteration of their inner membranes occurred. Maeno and others17 demonstrated that the earliest pathologic effect of artemisinin is on the parasitic membrane in P. falciparum, which seems to be the case in our study. We used a short treatment duration because we wanted to know if the artesunate-dihydroartemisinin combination could act as rapidly against T. gondii as observed against P. falciparum. Given this short period, the drugs show an interesting efficacy; however, some brain cysts in the cerebral tissue were still viable because they were able to produce secondary cysts in naive mice. We obtained a better inhibition of parasite growth results in vivo with our cyst-forming strain than with the RH strain, but in vitro it was lower than that previously observed by other.
