We have recently demonstrated that, in chronic smokers, cough reflex sensitivity is significantly enhanced as soon as two weeks after smoking cessation Eur Respir J 2005; 26: 481s ; , suggesting that diminished cough sensitivity in smokers results from chronic cigarette smoke-induced desensitization of airway cough receptors. In the present study, we measured cough reflex sensitivity to inhaled capsaicin C5 ; in 11 chronic smokers while they were smoking baseline study ; , after smoking cessation, and again after resumption of smoking. Two weeks after smoking cessation, there was a significant enhancement of cough reflex sensitivity; mean SEM ; log C5 decreased from 1.770.18 to 1.470.14 p 0.01 ; . All subjects resumed smoking after 2-12 weeks of abstinence and underwent repeat capsaicin challenge testing 14-23 days after resumption of smoking. Mean log C5 increased after smoking resumption compared to the last value obtained during the smoking cessation period: 1.770.16 vs.1.420.15 p 0.0004 ; . Anyway Rick says that for 50 minutes to an hour of fluoroscopy during an ablation a patient would receive approximately 100-200 millirems, possibly more if you are a "big" person. I believe that people working with radiation are allowed more mSv simply because they are around radiation equipment and will be exposed to more radiation than the average person. Since the virus enters the body via the enteric route, a strong local immune response that would diminish the infection at the site of entrance, is a desirable goal for vaccination. The gut associated lymphoid tissue GALT ; constitutes the lymphoid tissue of the intestines. This tissue includes Peyer's patches, which are specialized in uptake of pathogens and foreign materials through the M-cells. Antigens can be targeted to the Mcells by incorporation in particles with sizes up to 10 micrometer. Chitosan is a suitable material for particles, as it is non-toxic and biocompatible. The objective of this project was to visualize gene expression after incorporation of DNA into chitosan microparticles and oral delivery in cats. To enable visualization in vivo, the gene encoding the lacZ-gene was used. The lacZ gene was cloned into the vector pVR1012, resulting in pVR1012lacZ, and lacZ was successfully expressed in COS-7 cells. pVR1012lacZ was thereafter complexed to chitosan. Complexes were characterized regarding uniformity in size and morphology and stability under physiological conditions. Size measurements of particles were also performed. It was found that it is possible to prepare chitosan pVR1012lacZ complexes, which are stable at physiological pH and temperature, and have a size of about 250 nm. Treatment with chitinase, a natural occurring chitosan degrading enzyme, however disrupts the complexes. An in vivo experiment was performed to investigate whether complexes would reach and be taken up by the epithelial cells in the intestine when distributed orally to cats. X-gal staining of cat intestines 48 hours post delivery of complexes showed a significant increase in the expression of lacZ compared to background. These results identify chitosan DNA complexes as a promising candidate for an oral vaccine delivery system in cats.
Based on a series of this work, we were able to establish the general relationship between the structure of radicals formed and the polymer structural features. Crosslinkable polymers give the midchain alkyl radical as a major component radical, while degradable polymers give the end-chain alkyl radical as a major component radical, and if polymers are of branched structure or contain the aromatic ring, the crosslink reactions occur preferentially on these moieties. And, one of the common features is that dangling bond sites DBSs ; are more or less formed in all plasma-irradiated polymers resulting from the occurrence of CASING crosslinking by activated inert gas ; . DRUG ENGINEERING FOR DDS PREPARATION BY PLASMA TECHNIQUES For the most suitable therapy, development of sustained- and controlled-release systems for drug delivery is one of the most active areas today in the entire field of drug research. A wide variety of approaches to controlled-release DDSs have been thus far investigated for oral application. Oral drug delivery is the most desirable and preferred method of administrating therapeutic agents for their systematic effects such as patient acceptance, convenience in administration, and cost-effective manufacturing. We have developed plasma-assisted preparation of multilayered tablets applicable to an oral DDS. Figure 4 illustrates the schematic representation for preparation of double-compressed tablets and a drug dissolution test including the experimental set-up for plasma irradiation on the tablets.
