It has been reported that clotrimazole blocks maxi-K channels in smooth muscle cells Rittenhouse et al., 1997 ; . To address this question of specificity, we tested 300 nM clotrimazole on parotid maxi-K channels in IK1 ; animals. This concentration blocked only 4.1 2.1% N 7 ; of the maxi-K current measured at + 50 mV, confirming the IK1 specificity of this compound in our cells. If, as suggested above, an increase in IK1 activity leads to a reduced maxi-K activity, then there should be an inverse relation between these two. Thus we measured IK1 activity by the conductance at -110 mV normalized by cell capacitance ; and the maxi-K channel activity as the time-dependent conductance at + 50 many cells patched with various Ca 2 + concentrations. The pooled results of these experiments are illustrated in Fig. 2. It seems clear from these data that cells patched with higher intracellular Ca2 + generally resulted in larger IK1 conductance levels and, as predicted, smaller maxi-K levels and diclofenac.

Prominent among systemic factors predisposing to superficial candidiasis are diabetes mellitus, HIV infection and treatment with broadspectrum antibiotics or supraphysiological doses of adrenal corticosteroids. Women in the third trimester of pregnancy are prone to vulvovaginal candidiasis. Factors predisposing to deep candidiasis include immunosuppression including that due to HIV infection ; , indwelling intravenous catheters, neutropenia, hematological malignancies, burns, postoperative complications and very low birthweight in neonates. Furthermore, our data show that the inhibitor miconazole almost completely restored a normal response to the amino acid after nNOS blockade, suggesting a major role for a nNOS-related calmodulin-dependent cytochrome c reductase activity that interferes with increased arginase activity. This proposal is strongly supported by the demonstration of the presence of arginase II different from the liver cytosolic isozyme ; in the mitochondrial matrix of most tissues 56 ; , where the enzyme is involved in the production of ornithine as a precursor of glutamate, which might enter the TCA cycle after further metabolism to succinyl CoA.
